This Standard sets out a method for enumerating spores of sulfite-reducing clostridia andC.perfringens in water, using membrane filtration, with a confirmation procedure foridentifying C.perfringens.Refer to AS 4276.1 for a description of the general procedure for determining counts ofmicroorganisms by membrane filtration techniques.NOTES:1 Membrane filtration is suitable for enumerating microorganisms only when the turbidity ofthe water is low.2 The membrane filtration method is not suitable for waters containing only small numbers ofC.perfringens in the presence of many other species that are capable of growth on the mediumused.3 Bacterial counts derived from membranes with fewer than 20 or more than 80 colonies areapproximate.4 A flow diagram of the procedure is shown in Appendix A.
